Quantitative Cross-Linking of Proteins and Protein Complexes

Abstract Cross-linking, in general, involves the covalent linkage of two amino acid residues proteins or protein complexes close proximity. Mass spectrometry and computational analysis are then applied to identify formed deduce structural information such as distance restraints. Quantitative cross-linking coupled with mass is well suited study dynamics conformations complexes. The quantitative workflow described here based on application isotope labelled cross-linkers. Proteins present different states differentially cross-linked using a “light” “heavy” cross-linker. intensity ratios cross-links (i.e., light/heavy heavy/light) indicate changes interactions that maintained states. These insights lead better understanding function investigated. applicable wide range research questions including, for instance, upon ligand binding.
